چکیده

Thermophilic mannanase from the Bacillus sp. MG-33 was applied on wheat straw-rich-soda pulp to evaluate its bleaching potential. Optimum conditions for bio-bleaching of pulp were found as, mannanase 3Ug -1 of pulp at pH 6.5 and temperature 65oC that enhanced the brightness by 30% and reduced the kappa number by 20% within 2h. Tear index (1.1, points) and burst index (0.02, points) were also improved by mannanase-treated pulp as compared to the untreated pulp. Treatment of chemically (CEHIHII) bleached pulp with an enzyme showed significant effect on release of chromophores, hydrophobic and reducing compounds. Mannanase-prebleaching of raw pulp reduced the use of hypochlorite by 28% to achieve brightness of resultant hand sheets similar to the fully chemically bleached pulp. Scaning electron microscopic studies of mannanase treated pulp distinctly revealed that separation of microfibers which assist in lowering down the chemical requirement in the subsequent chemical bleaching.

SOLID STATE FERMENTATION AND CHARACTERIZATION OF PARTIALLY PURIFIED THERMOSTABLE MANNANASE FROM Bacillus sp. MG-33

Bacillus sp. MG-33 was isolated from the desert of Rajasthan (India). The organism produced 500 and 200 Ug of thermostable mannanase (after 96h) in solid state fermentation (SSF) of wheat bran and wheat straw rich-soda pulp at the moisture ratio of 1:1.5 and 1:3 at 30oC, respectively. Two-step partially purified mannanase was optimally active at 65oC and was 100% thermostable at 55 to 60oC for ...

Biobleaching of Wheat Straw Pulp using Laccase and Xylanase

Wheat straw pulp can be considered to be one of most important raw materials for specialty and functional paper products. Bleaching sequences involving laccase and xylanase were applied to bleach wheat straw pulp. The bleached pulp properties after a sequence of xylanase (X), laccase (L), and extraction (E), i.e. XLE, were compared with those of LE and LEX sequences. It was found that the XLE b...

Microbial diversity during composting cycles of rice straw

Mesophilic bacteria, fungi and actinomycetes varied during composting cycles with high numbers in the initial and final cycles with maximal values in compost C and D, and sharply decreased in the heating cycles. Staphylococcus aureus and Bacillus subtilis were dominated at the initial composting cycle. Whereas, at maturity Bacillus subtilis was the major followed by B.badi...
